摘要: To extract total RNA from biomass is an important experimental technology frequently used in modern molecular biology research. The three method of guanidine thiocyanate, STE and Trizol reagent were used to extract total RNA from flammulina velutipes. The operation processes of the three methods were described in detail. Formaldehyde agarose gel electrophoresis was used to detect the total RNA. The results show that all the three methods are suitable for the molecular biology research related to space flammulina velutipes, and guanidine thiocyanate method is the best one for the genome RNA extraction from space flammulina velutipes. With suitable precipitation time and sample weight (5 ul), more and purer total RNA were extracted and the ratio of 28s/18s electrophoresis brightness was about 2: 1. The extraction of total RNA is of great significance for the subsequent molecular experimental study on RT-pcr, gene library construction, etc.
